Study Hints That Statins Might Fight Breast Cancer (HealthDay)

THURSDAY, Jan. 19 (HealthDay News) -- Amid hints that statins -- cholesterol-lowering drugs -- might also play a role in preventing or treating certain types of cancer, new research sheds some light on how these drugs may help stop breast cancer in its tracks among certain women.

The p53 tumor suppressor gene stops the uncontrolled growth of cancer cells, but some women with breast cancer have mutant forms of this gene. In the new study, when the mutant p53 cells were treated in the laboratory with statins, the cells stopped their erratic growth, and even died in some cases.

It seems that the mutated p53 genes may activate the same pathway that the statins inhibit -- the mevalonate pathway, the study suggests. The mevalonate pathway is important in the body's production of cholesterol.

In the study, the effects of the statin drugs were erased when the mevalonate pathway was reactivated, supporting the potential mechanism. The new research is published in the Jan. 20 issue of the journal Cell.

Study author Dr. Carol Prives, chair of the department of biological sciences at Columbia University in New York City, is cautious in her enthusiasm about the results and their implications.

"The study is adding the possibility that there may be classes of breast cancer patients who will respond better to statins than others," she said, but noted that this research is far away from the bedside.

"By understanding better what sort of cells would respond to statins, one might have a better idea of whether or not to consider using them," she added. "The next step could be a trial of statins in women with breast cancer who have a mutated copy of the p53 gene."

Commenting on the study, cancer expert Marc Symons said, "This paper unravels a mechanism whereby p53, a frequently mutated cancer gene, promotes the aberrant behavior of cancer cells."

The mutated protein stimulates the mevalonate pathway, explained Symons, an investigator at the Center for Oncology and Cell Biology at the Feinstein Institute for Medical Research in Manhasset, N.Y.

"Statins, drugs that are widely used to lower cholesterol levels, block a key step in the mevalonate pathway," Symons said. "The new results may well give new momentum to the use of statins as anti-cancer agents."

Dr. Stephanie Bernik, chief of surgical oncology at Lenox Hill Hospital in New York City, is also intrigued by the potential of the new findings.

"This paper addresses a possible new target for therapeutic agents based on a well-known tumor suppressor gene that is common in many cancers," Bernik said. "Identifying novel pathways that lead to tumor formation is the first step to developing new drugs that can specifically target some of the complex mechanisms that contribute to the development of cancer," she pointed out.

"This work and other projects like this raise the hope that we will one day be able to cure cancers on a molecular level," Bernik said.

Falcons hire Mike Nolan to lead defense (AP)

ATLANTA ? The Atlanta Falcons hired former San Francisco coach Mike Nolan as their defensive coordinator on Wednesday, hoping he can toughen up a unit that has rarely ranked among the NFL elite.

Nolan spent the last two seasons as Miami's defensive coordinator. Prior to that, he served four years as the 49ers head coach, following in the footsteps of his father.

In all, Nolan has been an NFL coordinator for 14 years. He takes over in Atlanta for Brian VanGorder, who returned to the college ranks to take the same position at Auburn.

The Falcons will have two new coordinators next season. Dirk Koetter was hired Sunday to run the offense after Mike Mularkey left to become head coach of the Jacksonville Jaguars.

In another move toward revamping the staff for 2012, head coach Mike Smith announced that offensive line coach Paul Boudreau has been let go. The Falcons line struggled this season, most notably in an ugly 24-2 loss to the New York Giants in the opening round of the playoffs ? a game in which Atlanta twice failed to convert on fourth-down quarterback sneaks by Matt Ryan.

The hiring of Nolan figures to be met with more enthusiasm in Atlanta than Smith bringing in Koetter to run an offense that has plenty of weapons but largely failed to meet expectations. Koetter had been the offensive coordinator in Jacksonville, which finished last in total offense and in yards passing.

Nolan's numbers are much more impressive. The Dolphins allowed the sixth-fewest points in the NFL (19.6 per game), ranked third in rushing yards (95.6) and tied for 10th with 41 sacks. The biggest question is how to mesh Nolan's preference for the 3-4 alignment with personnel that is built around Smith favoring the 4-3.

"Mike Nolan is an established coordinator in this league," Smith said in a statement. "Mike has a history of developing physically intimidating defensive units, and we are fortunate to have him join our staff."

The Falcons haven't ranked among the top 10 in yards allowed since 1998, their only Super Bowl season.

The 52-year-old Nolan didn't have much success as a head coach, going 18-37 with the 49ers ? a stint that was perhaps most noteworthy for his desire to wear a suit on the sideline in honor of his father, former 49ers and New Orleans Saints coach Dick Nolan. The league initially resisted, citing its apparel contracts, but eventually worked out a new arrangement.

Nolan is regarded as one of the NFL's top defensive minds. He was only 34 when tapped by former Falcons coach Dan Reeves to take over as the Giants defensive coordinator in 1993. He's also served as a coordinator with Washington, the New York Jets, Baltimore and Denver.

In Baltimore, Nolan served on the same staff with Smith.

Two attacks kill 13 in Afghan south (Reuters)

HELMAND, Afghanistan (Reuters) ? A suicide bomber and a remotely detonated mine killed 13 people including a security official on Wednesday in Afghanistan's southern Helmand province, an area where NATO-led forces have claimed security improvements over the past year.

The insurgent bomber detonated explosives strapped to his body in a crowded market in Helmand's Kajaki district in the afternoon, killing seven civilians and two Afghan policemen and wounding eight more, said governor spokesman Daoud Ahmadi.

Hours later, a mine killed a district security official and three others people in Nad Ali district, police chief Marjan Akmal told Reuters, declining to go into detail.

Both attacks raise serious concerns about security in one of the country's most violent provinces, an area that saw fierce fighting during a surge of U.S. troops over recent years.

So far nobody has claimed responsibility for the attacks, which come weeks after an offer from the Taliban to open a political office in Qatar as a prelude to possible peace talks with the United States and other nations.

British forces handed security of Kajaki district to U.S. Marines in June 2010, though the British remain the dominant foreign force in Helmand, a province that borders Kandahar, the birthplace of the Taliban and its main southern stronghold.

Despite the presence of more than 100,000 foreign troops, violence across Afghanistan remains at its worst levels since the Taliban were toppled by U.S.-backed Afghan forces in late 2001, according to the United Nations.

Foreign forces fighting mainly Taliban insurgents are in the process of handing control of security over to the Afghan army and police, with foreign combat troops due to leave by the end of 2014.

Leap second decision is postponed

A decision on whether to abolish the leap second - the occasional, extra second added to the world's time - has been deferred.

Experts at the International Telecommunication Union (ITU) were unable to reach a consensus, so moved the matter to a meeting in 2015.

The US argued at the meeting that leap seconds were causing problems for communication and navigation systems.

But the UK said that the long-term consequences of losing it were great.

An ITU spokesman said that Canada, Japan, Italy, Mexico and France all supported the United States' stance on losing the leap second, while Germany, like the UK, wanted the extra second to stay.

More countries though, including Nigeria, Russia and Turkey, wanted further study.

As a result, the ITU decided that more research was needed to consider the broader social implications of losing the leap second before a decision could be taken.

The ITU suggested that a study group should investigate the issue, before presenting any proposals at the next World Radio Conference in 2015.

It means that for now, the world's time will continue to be linked to the Earth's rotation.

The next leap second is due to be added on 30 June 2012.

Out of time

The leap second was introduced in 1972.

It was added to keep our modern timekeepers - atomic clocks, which rely on the vibrations in atoms to provide a very accurate measurement of time - in line with our slightly less reliable timekeeper, the Earth.

Because our planet wobbles a little on its axis as it spins, it means some days end up being a few milliseconds longer or shorter than others.

This means that over time, the time based on atomic clocks, and the time based on the Earth's rotation drift further and further apart.

When this difference is deemed by the International Earth Rotation Service, which monitors the Earth's activity, to be approximately 0.9 seconds, a leap second is added to pull the two back into sync.

Sometimes a leap second can be added every year, sometimes not for several, with six-months' notice provided before action needs to be taken.

What is a leap second? The BBC's Rebecca Morelle went to Greenwich's Royal Observatory to find out

Those who wanted to lose the leap second said that the one-second increments were becoming increasingly problematic for a vast range of modern navigational and communication systems, such as sat-nav, financial services, air traffic control and the internet.

These all rely on having a continuous and stable timescale, so adding a somewhat unpredictable, one-second increment can be disruptive.

The UK, though, says any problems are exaggerated - and that losing the leap second could cause long-term problems, as the time based on the atomic clocks and the time based on the Earth's rotation would move ever further apart.

Over decades, this would amount to a minute's difference, but over 500 years this could be an hour, and over thousands of years, the Sun could be setting when atomic clocks claim it is morning.

Peter Whibberley, senior research scientist at the National Physical Laboratory in Teddington, UK, who is leading the UK's delegation, said: "This result achieves the UK's aims of securing much broader debate and understanding of the consequences of ending the link between UTC (Coordinated Universal Time) and solar time before a final decision is taken."

Merkel returns to AC Milan after Boateng injury

Associated Press Sports

updated 4:41 p.m. ET Jan. 18, 2012

MILAN (AP) -AC Milan has brought back German midfielder Alexander Merkel on loan after losing Kevin-Prince Boateng for a month with a torn left thigh muscle.

Milan made the announcement on its website on Wednesday, saying the move is a temporary loan since Merkel's rights are jointly shared by Milan and Genoa.

The 19-year-old Merkel made 10 appearances and scored one goal in all competitions for Milan last season. During the first half of this season, he played 14 times for Genoa.

Milan announced Boateng's injury late Tuesday, adding that goalkeeper Christian Abbiati has a slight left calf problem.

Milan trails Serie A leader Juventus by one point.

Some Transformer Prime users unable to upgrade to ICS, serial number 'Unknown'
Ice Cream Sandwich for the Transformer Prime was one of the most anticipated updates for one of the most anticipated tablets of recent memory. It was pushed out last week and so everyone's happy -- right? Sadly, some folks are missing out. We've received a number of reports from users who haven't been as successful as we were in updating to Android 4.0, their slates showing an "Unknown" serial number preventing the application of the update. ASUS is looking into the issue and affected users were told to perform a factory reset and, if that doesn't work, try it again and let the slate sit at the desktop for about 15 minutes. Let us know how you're faring in comments, and hit up the xda developers links for more support should you need it.

[Thanks, Ben]
